Application Abstract: Diabetes-induced renal fibrosis(RF)serious harm to human health,the exact mechanism has not been fully elucidated.In the early and mid-RF process,PVT1 is the important regulatory factor that TGF-β1-induced renal cell type Ⅱ mesenchymal transition(EMT),and an important node in G6PD/ROS/p38MAPK pathway.We hypothesized that:PVT1 can effectively regulate G6PD/ROS/p38MAPK pathway,which will become a new mechanism of RF.Previous studies found that:Based on the Hui medicine"Four Xing"theory,Hui-hui Gan-song Yin(HGY)is effective in improving clinical symptoms of patients with diabetic nephropathy;and lower blood sugar and blood lipid in DM rats.To clarify HGY’s efficacy and mechanism in the early-mid RF,this study intends to introduce KKAy mouse model.The use of molecular biology,confocal laser technology and tools,from animals-cells-molecular level to putting PVT1 as the main target,observing HGY regulate G6PD/ROS/p38MAPK pathway in mice as early-mid RF and podocyte cell EMT mechanism.
