项目标书摘要:“藏羊多胎性能选育及杂交改良研究与示范”科技报告摘要 2014年青海省科技厅下达第二批新开基本科技计划“藏羊多胎性能选育及杂交改良研究与示范”项目[2014]83号,合同执行期2014~2017年,申请延期一年。项目由青海省畜牧兽医科学院主持,青海省海北州畜牧兽医科学研究所、祁连县祁连山藏羊种羊繁育场、中国农业科学院兰州畜牧与兽药研究所、山东省农业科学院畜牧兽医研究所共同参与完成。藏羊多胎连续繁殖能力研究共调查羊群111群,发现52.25%的羊群中有繁殖双羔的母羊,多胎母羊占调查母羊总数的0.90%。在此基础上将分散的个体集中组建多胎母羊群,存栏一代母羊251只,繁育二代母羊存栏612,三代母羊156只,合计存栏多胎母羊达到1019只。检测藏羊多胎FecB基因1232只,结果证明藏羊中存在BMPR IB基因FecB突变,并且成功选育16只纯合子种公羊已经投入使用。连续多胎繁殖能力研究一代母羊集中后第一年有38.46%的母羊连续产双羔,繁殖率136.36%,第二年经过选育和加强饲养管理双羔率提高到51.56%,繁殖率达到151.60%。二代母羊产第三胎时繁殖率达到152.63%,双羔率54.66%,表明藏羊多胎繁殖性能能够遗传给下一代,其次双羔率也与胎次、饲养管理等具有重要关系。小尾寒羊与藏羊杂交培育多胎羊试验研究课题引进纯合子GG型FecB基因小尾寒羊种公羊12只,组建母羊群规模500只。研究小尾寒羊引进到高海拔严寒地区生态适应性良好,血红蛋白含量随海拔升高而逐渐升高。种公羊能够正常生长发育和采精、配种,平均采精量0.97ml,2~2.5岁成年时,平均采精量1.6ml,精子的活率和活力显著提高。繁活寒藏F1代羔羊1504只,平均繁活率85.45%。其中选留种公羔40只,母羔669只,出栏775只。寒藏F1代初产母羊横交产羔172只,繁殖率135.33%,其中产双羔母羊46只,双羔率36.5%。第二胎产羔157只,繁殖率152.43%。寒藏F1代初生重公羔4.63kg,母羔4.61kg,分别比藏羊高18.71%和24.26%。周岁寒藏F1代与藏羊公羊平均体重分别为37.49kg和30.98kg,母羊分别为38.96kg和30.81kg,寒藏F1代公、母羊分别比藏羊高26.46%和21.01%。对寒藏和藏寒杂交羊进行FecB基因检测,寒藏F1代存在GG型(纯合型)、GA(杂合型)和AA野生型)三种基因型,基因型频率分别为0.03、0.79和0.18;藏寒F1代GA和AA基因型的频率分别为0.81和0.19。FecB基因能遗传给杂交后代,这为利用FecB基因进行多胎分子标记培育新多胎新品种提供了依据。发表论文7篇,编制规范1项,授权实用新型专利1件,新增利润40.79万元。新增就业 7人。完成或超额完成了项目合同规定的任务内容和指标。关键词:藏羊多胎性能选育、双羔、BMPR IB、FecB基因、小尾寒羊与藏羊杂交改良、羊多胎基因导入、小尾寒羊血液生理指标、小尾寒羊精液品质
Application Abstract: Research and demonstration on Multi-birth performance selection and hybrid improvement of Tibetan sheepSummary of scientific report In 2014,the China Qinghai Provincial Science&Technology Department issued the second batch of new basic science and technology plan"Tibetan sheep multiple embryo performance breeding and hybrid improvement research and demonstration"project[2014]no.83.The execution period of the contract is from 2014 to 2017,which was applied for a one-year extension.The project is hosted by Qinghai Provincial College of Animal Husbandry and Veterinary Medicine,with the participation of Haibei Institute of Animal Husbandry and Veterinary Medicine,Qilian county Qilian mountain Tibetan sheep breeding farm,Lanzhou Institute of Husbandry and Pharmaceutical Sciences,Chinese Academy of Agricultural Sciences,and Institute of Animal Sciences and Veterinary Medicine,Shandong Academy of Agricultural Sciences.A total of 111 groups were investigated in the study of continuous reproductive ability of multiple foetuses of Tibetan sheep.It was found that 52.25%of the sheep had female sheep with twin lambs,multiple ewes accounted for 0.90%of the total number of ewes surveyed.On this basis,the scattered individuals are concentrated to form a multi-litter ewes,there are 251 ewes of the first generation,612 second-generation ewes and 156 third-generation ewes in stock,and the total number of multiple ewes in stock has reached 1019.1232 Tibetan sheep were tested for multiple FecB gene,the results showed that BMPR IB gene FecB mutation was present in Tibetan sheep,and the successful breeding of 16 homozygote rams has been put into use.Study on the reproductive capacity of the first generation of ewe showed that 38.46%of the ewes produced two lambs continuously in the first year,and the reproduction rate was 136.36%.In the second year,the rate of double lambs was raised to 51.56%and the reproduction rate was 151.60%after breeding and strengthening the feeding management.The reproduction rate of the second generation ewes reached 152.63%and the twin lambs rate 54.66%when they gave birth to the third baby.The results showed that the reproductive performance of Tibetan sheep could be passed on to the next generation,and the rate of twin lambs was also related to litter size,feeding management and so on.12 male sheep of small tail Han sheep with GG type FecB gene homozygote were introduced in the experimental study on Multifetal Sheep breeding between small tail Han Sheep and Tibetan Sheep,and a herd of 500 sheep were set up.The small tail Han sheep was introduced to the cold area at high altitude and the ecological adaptability was good,and the hemoglobin content increased gradually with the increase of altitude.The male sheep was able to grow and develop normally and collect sperm,and the average sperm intake was 0.97 ml,average sperm intake of 2-2.5-year-old adult rams were 1.6ml,sperm viability and motility were significantly increased.1504 lambs of F_1 generation were bred in Hanzang,with an average survival rate of 85.45%.Among them,40 male lambs and 669 female lambs were selected for breeding,775 sheep were sold.172 lambs were crossbred in Han Zang F1 generation,and the reproduction rate was 135.33%.Among them,46 ewes gave birth to double lambs,and the rate of double lambs was 36.5%.157 lambs were born at the second birth,and the reproduction rate was 152.43%.The birth weight of male lambs and female lambs in Han Zang F1 generation were 4.63 kg and 4.61 kg,which were 18.71%and 24.26%higher than those of Tibetan sheep,respectively.The average weight of one year old Han-Zang F1 ramand Tibetan ram were 37.49kg and 30.98kg respectively,and that of ewe were 30.81kg and 30.81kg,respectively.The average weight of Han-Zang F1 generation rams and ewes were 26.46%and 21.01%higher than that of Tibetan sheep,respectively.The detection of FecB gene in Han Zang and Zang Han hybrid sheep showed that there were three genotypes of GG genotype(homozygous),GA(heterozygote)and AA wild type in Han Zang F1 generation,the genotype frequencies were 0.03,0.79 and 0.18,respectively.The frequencies of GA and AA genotypes in Zang Han F1 generation were 0.81 and 0.19,respectively.The FecB gene can be passed on to hybrids,which provides a basis for the use of FecB gene for multi-embryo molecular markers to develop new multi-embryo varieties.Published 7 papers,compiled 1 specification,authorized 1 utility model patent,increased profit by 407,900 yuan.Seven new jobs were created.Complete or overfulfill the tasks and targets specified in the project contract.Key words:Tibetan sheep multiple fetus gene,double lambs,BMPR IB gene,FecB gene,sheep multiple performance breeding,sheep multiple embryo gene introduction,small tail Han sheep blood physiological index,small tail Han sheep and Tibetan sheep hybridization,semen quality of small tail Han sheep
