项目标书摘要:本项目在前期积累的大量直突摇蚊亚科(Orthocladiinae)蛹期标本基础上结合中国研究材料上的优势,通过多方合作和广泛采集,利用比较分类学和DNA条形码技术,对中国直突摇蚊亚科蛹期进行分类学研究。研究旨在规范国内分布的直突摇蚊亚科族级、属群和属级蛹期鉴别特征,核查并订正国内直突摇蚊亚科蛹期已知种类,纠正部分物种的错误鉴定,补充和完善种类描述和特征图,完善中国直突摇蚊亚科习见种的不同虫态“配套”研究。整理中国直突摇蚊亚科种类名录,提供修订后的国内直突摇蚊亚科蛹期不同阶元检索表、蛹期鉴定手册等分类资料,为制定合理的摇蚊科分类体系奠定基础,还可为世界摇蚊研究工作提供中国地区生物多样性资料。此外,研究结果也对害虫防治、水环境质量评价和水产养殖具有重要的理论价值和实践意义。

Application Abstract: Based on accumulation of pupal materials of Orthocladiinae and the advantage of Chinese materials,the project will combine the method of comparative morphology with DNA barcoding to study the taxonomic revision on pupal stage of Orthocladiinae through multilateral cooperation and extensive collection.The research will revise the diagnostic characters of tribes,generic groups and genera on pupal stage of Orthocladiinae from China,systematically organize and revise the materials of identification in pupal stage of Chinese Orthocladiinae.The description and characteristic figures of known species and the research of all life stages of common species will be supplemented and perfected.Catalogue of Chinese Orthocladiinae,revised keys of known pupal stage of Chinese Orthocladiinae on different level taxa and manual of pupal stages identification will be given.The research achievement will lay a foundation for reasonable classification system of Chironomidae and provide the biodiversity information of chironomids from China for the world.In addition,and it will have great value in theory and practical significance for pest control,water quality assessment and aquaculture.

项目基于形态特征和分子数据对中国直突摇蚊亚科(Orthocladiinae)蛹期进行分类学研究。主要开展的研究内容是:1)直突摇蚊亚科蛹期鉴定、生活史“配套”;2)种类修订、近缘种区分;3)分类学资料系统整理。通过研究整理和记述直突摇蚊亚科蛹期30属126种,整理了直突摇蚊亚科不同阶元(分属、种)检索表等分类资料,补充直突摇蚊亚科物种记录。基于形态学和分子数据,完成了Epoicocladius,Boreoheptagyia,Limnophyes等属级修订,报道新种7个,新纪录属1个,新纪录种4个,同物异名1个。同时,基于条形码数据发现14个未定名种和1个潜在同物异名。基于试验材料提取和上传了74种180余条直突摇蚊亚科条形码数据。针对北方习见摇蚊红裸须摇蚊Propsilocerus akamusi开展了基因组学和转录组学研究,研究公布了摇蚊科中首个染色体水平上的高质量基因组数据—红裸须摇蚊基因组。相关研究结果进一步丰富了中国摇蚊物种生物多样性,基础研究资料在水环境监测和河流健康性评估方面得到了应用。

    Glutathione S-transferases (GSTs) are major enzymes in detoxification phase II, and have been functioned in resistance to various insecticides or oxidative stress. Herein, we selected the non-biting midge, Propsilocerus akamusi, widespread in Asian aquatic ecosystems, to uncover the gene location, structure, and phylogenetics relationship of GSTs at genome scale first time. Thirty-three cytosolic and four microsomal GST genes were identified and located on the four chromosomes. The cytosolic GSTs involved in the eight subclasses and five GST genes were unclassified. The expansion of GST genes in P. akamusi experienced duplication events on the delta, theta, xi, iota, and unclassified subclasses. The RNA-Seq analyses and RT-qPCR validation showed that the expression of PaGSTt2 gene is significantly elevated, with deltamethrin concentration increasing. The tertiary structure of PaGSTt2 enzyme was reconstructed, which was different from the other theta gene in the active site. In addition, the GST genes of six chironomids were first described based on the assembled genomes to explore the difference of those in the adaptation to kinds of environments. The GST frame for P. akmusi and its expression profiles provide valuable resources to understand their role in insecticide resistance of this species, as well as those of other biting midges.

    The genus Parachironomus has a cosmopolitan distribution including 85 valid described species world-wide. Species records and studies of the genus in the Tibetan Plateau are scarce. In this study, the genus Parachironomus from China is revised and two new species, Parachironomus wangi Liu & Lin, sp. nov. and Parachironomus nankaiensis Liu & Lin, sp. nov., are described based on adult morphology and molecular data. Paracladopelma demissum Yan, Wang & Bu is placed in the genus Parachironomus as a new combina-tion. A neighbor-joining tree was reconstructed based on all known Parachironomus COI DNA barcodes. A key to adult males of the genus Parachironomus from China is also provided.

    The male adult of Boreoheptagyia zhengi Lin & Liu, sp. nov. is described and illustrated based on material collected in China. Associated morphological characteristics and reference to its DNA barcode are provided. Boreoheptagyia kurobebrevis (Sasa & Okazawa, 1992) is newly recorded from China based on both a male and female, with additional associated data on the DNA barcode of the male adult.   • 10.Toward a global DNA barcode reference library of the intolerant nonbiting midge genus Rheocricotopus Brundin, 1956

    Environmental DNA metabarcoding is becoming a predominant tool in biodiversity assessment, as this time- and cost-efficient tactics have the ability to increase monitoring accuracy. As a worldwide distributed genus, Rheocricotopus Brundin, 1956 still does not possess a complete and comprehensive global DNA barcode reference library for biodiversity monitoring. In the present study, we compiled a cytochrome c oxidase subunit 1 (COI) DNA barcode library of Rheocricotopus with 434 barcodes around the world, including 121 newly generated DNA barcodes of 32 morphospecies and 313 public barcodes. Automatic Barcode Gap Discovery (ABGD) was applied on the 434 COI barcodes to provide a comparison between the operational taxonomic units (OTU) number calculated from the Barcode Index Number (BIN) with the "Barcode Gap Analysis" and neighbor-joining (NJ) tree analysis. Consequently, these 434 COI barcodes were clustered into 78 BINs, including 42 new BINs. ABGD yielded 51 OTUs with a prior intraspecific divergence of Pmax = 7.17%, while NJ tree revealed 52 well-separated clades. Conservatively, 14 unknown species and one potential synonym were uncovered with reference to COI DNA barcodes. Besides, based on our ecological analysis, we discovered that annual mean temperature and annual precipitation could be considered as key factors associated with distribution of certain members from this genus. Our global DNA barcode reference library of Rheocricotopus provides one fundamental database for accurate species delimitation in Chironomidae taxonomy and facilitates the biodiversity monitoring of aquatic biota.
