The non-selectivity and undesirable water solubility are major limitations for the utilization of anti-cancer drugs in traditional therapy, leading to diminished therapeutic efficacy and severe side effects. Besides, the tumor inhibitory effect of photodynamic therapy (PDT) is limited, due to the limited depth of light penetration. However, combination therapies can reverse the dilemma that monotherapies face in clinical use. Here, a stimulus-sensitive drug delivery system was prepared by self-assembly for synchronized delivery and combination therapy. It was constructed by employing pH-responsive imine bonds to attach the chemotherapeutic drug daunorubicin (DNR) and the photosensitizer methyl aminolevulinate (MAL) to oxidized hyaluronic acid (OHA), named NPs(MAL/DNR). The nanoparticles demonstrated excellent inhibitory effect and synergistic effect in tumor suppression, as evidenced by in vitro cytotoxicity results (CI = 0.90, synergism). In summary, the prepared dual-drug nanoparticles can play a synergistic role in selectively killing tumor cells. This provides a new feasible direction for the use of combined chemotherapy and photodynamic therapy in the treatment of breast cancer. © 2025 Elsevier B.V.

A glutathione (GSH)-responsive nano -drug delivery system combining photodynamic therapy (PDT) and chemotherapy (CT) was developed for lymphoma. The small -molecule reduction -sensitive 6-mercaptopurine (PTA) and photosensitizer chlorine6 (Ce6) were loaded onto carboxymethyl chitosan (CMCS), a biocompatible and water-soluble carrier. Through ultrasonic self -assembly, dual drug combination nanoparticles (CCP NPs) with an average particle size of 169 +/- 1.9 nm in the water have been developed. PTA is sensitive to GSH and degrades quickly in tumor cells with high GSH levels, which causes the disintegration of nanoparticles and the release of 6-mercaptopurine (6 -MP). Immediately thereafter, the Ce6 embedded in the nanoparticles was released to achieve sequential drug release. The PTA structure's Michael receptor significantly reduces GSH levels in tumor cells, which diminishes GSH's antioxidant influence on photosensitizers' ability to produce reactive oxygen species (ROS) and enhances PDT in a synergistic way. The CCP NPs revealed a nanometric size distribution, optimal surface charge and excellent stability. An in vitro toxicity study revealed that 6 -MP and Ce6 exhibited strong selectivity (SI = 4.23) and a synergistic impact (CI = 0.23) on Black Burkitt's lymphoma cells (Raji cells). Research on drug delivery systems that combine Ce6 and 6 -MP is a valuable new strategy for lymphoma.

Sometimes traditional hydrogel dressings for wound repair were difficult to meet the needs of use due to insufficient adhesive, antibacterial and antioxidant properties, so a high-performance and multifunctional poly(vinyl alcohol)-based hydrogel dressing was prepared using poly (vinyl alcohol) (PVA), polydopamine (PDA), and tannic acid (TA) for dual adhesive, antibacterial, and antioxidant properties in this study. The structure and morphology of the hydrogel were analyzed by FTIR and SEM. The effects of the concentrations of PVA, PDA and TA on the swelling, antioxidation and adhesion of the hydrogel were investigated. The results showed that the maximum equilibrium swelling of the hydrogels were all below 6 %, and they had good anti-swelling effects in different solution environments. The maximum skin adhesive strength of the hydrogels was 75.7 kPa, and the scavenging efficiencies against free radicals were above 90 %. Due to the inherent bacteriostatic ability of TA and the good photothermal conversion efficiency of PDA, the hydrogels could inhibit E. coli and S. aureus up to 99 %. The hydrogels possessed excellent dual antioxidant, antibacterial and adhesive properties, which greatly broadened the applications of PVA composite hydrogels. © 2023 Elsevier Ltd

以异氰酸酯和聚醚多元醇为主要原料,水、二氯甲烷、三乙烯二胺、辛酸亚锡和硅油作为添加剂,氨基酸溶液作为抗菌剂,通过一步发泡法制备了氨基酸改性的聚氨酯泡沫,并使用聚多巴胺/碳纳米管对聚氨酯泡沫进行表面涂覆,得到一种新型抗菌、吸油聚氨酯泡沫,并研究了其结构、形貌和性能。结果显示,该泡沫具有三维网状结构,与改性前的泡沫相比,其断裂强度提高至1.9倍,水接触角由3.28°提高至153.25°,能够吸附自身质量23.46倍的油类;经过10次循环使用,仍能保持最初吸油量的96.3%,抗菌效果达到了88.6%。

The fabrication of bio tissues using anisotropic constructs capable of repairing and/or replacing diseased bio tissues remains a significant challenge in tissue engineering. Therefore, this study investigated the fabrication of an anisotropic PLGA-microspheres/PVA-hydrogel composite using a novel directional freezing-thawing (DFT) technique. The DFT technique altered the properties of the anisotropic PLGA-microspheres/PVA-hydrogel composite, such that its compressive strength improved from 14.64 ± 1.09 MPa after three DFT cycles to 45.77 ± 6.73 MPa after nine cycles. The utilization of the DFT technique was also shown to enhance the proliferation and adhesion of cultured chondrocyte cells. Skin trauma is a widespread, extremely susceptible health issue that affects people all over the world. In this study, an innovative antibacterial hydrogel (ODAA hydrogel) with photosensitizer and antibiotics was developed. Oxidized dextran (ODEX) was used as a carrier to prepare a pH-responsive hydrogel by loading the antibiotic amikacin (AMK) and the photosensitizer hexyl 5-aminolevulinate (HAL) via imine bonds. The ODAA hydrogel has a uniformly distributed cavity structure. The cumulative release rates of HAL and AMK in a simulated inflammatory environment at pH 5.0 were approximately 62.3 % and 71.9 % during 15 days. These results demonstrate the ODAA hydrogel's ability to deliver antibiotics on demand, where the antibiotic content is reduced within the effective range. Regarding the in vitro antibacterial behavior, the combination of HAL and AMK synergistically destroyed the majority of Gram-positive and Gram-negative bacteria through several pathways with broad-spectrum antibacterial effects. ODAA hydrogel has been shown to be biocompatible, nearly non-cytotoxic, and capable of promoting wound healing. It is anticipated that the simultaneous targeted delivery of multiple drugs to lesions in the same carrier at ideal dose ratios for particular therapeutic combinations will produce the most synergistic effects. © 2023

To enhance bacteria-targeting and improve bactericidal activity, a novel GSH-responsive photodynamic anti-microbial nanosystem based on Alginate oligosaccharide-protoporphyrin IX (AOS-ss-PpIX) conjugates were constructed. The fluorescence self-quenching caused by the aggregation of PpIX groups into hydrophobic core could promote the photostability of PpIX and avoid the phototoxicity of the non-infected sites of the target plants. But once exposed to GSH stimulus, the recovery of high photodynamic activity were observed due to the destruction of the system and accompanied by the release of thiolated PpIX, and the photodynamic activity after 2 h was higher than that of PpIX under the same conditions. Furthermore the system showed more efficient photodynamic antibacterial capacity under light exposure. Moreover, the pot experiment also verified that the system showed outstanding control efficacy against cabbage soft rot caused by Erwinia carotovora Sclerotinia. Safety test exhibited that the size of nanoparticles can restrict the entry of nanoparticles into the plant cells to trigger photocytotoxicity. The design strategy provides a platform for promoting the utilization of photoactivated pesticides and reducing or avoiding their harm to target plants and the environment.

利用丝素肽对羧丁酰壳聚糖进行接枝改性,再以氧化魔芋(OKGM)作交联剂,通过Schiff碱反应制备得到一种可注射的羧丁酰壳聚糖-丝素肽/氧化魔芋(NSGM)水凝胶。样品的红外光谱和磁共振氢谱证实了改性、接枝和交联反应的发生,扫描电镜结果表明该水凝胶具有内部连通的孔结构。性能研究结果表明,该水凝胶具有35~70 s可调节的凝胶时间,溶胀率最高可达1 259%,室温下24 h后最多可维持21.1%的水分,同时具有良好的生物相容性。

Scars are consequences of the wound healing process, and eliminating scar formation remains a significant challenge. Here, an injectable HMSC hydrogel was developed based on silk fibroin peptide grafted hydroxypropyl chitosan (HPCS-g-SFP) and oxidized microcrystalline cellulose (OMCC) via Schiff base bonds. The synthesized HPCS-g-SFP copolymer displayed efficient free radical scavenging ability on hydrogen hydroxyl radicals and 1,1-diphenyl-2-picrylhydrazyl radicals (DPPH). The pore size, gelling time, equilibrium swelling rate and water retention properties of HMSC hydrogel could be regulated by changing ratio of OMCC and HPCS-g-SFP. Then, tetramethylpyrazine (TMP) was encapsulated into HMSC hydrogel to obtain TMP-loaded HMSC hydrogel. The TMP-loaded HMSC hydrogel facilitated 95% cell activity retention after culturing with human skin fibroblasts (HSF) or human hypertrophic scar fibroblast (HSFB) cells for 24 h. Additionally, in vivo animal experiments confirmed that TMP-loaded HMSC hydrogel promoted rapid wound healing while preventing scar formation. The designed injectable TMP-loaded HMSC hydrogel has potentials in promoting scarless wound healing.

A simple method had been developed through O-chitosan quaternary ammonium salt (O-HACC), polyvinyl alcohol (PVA) and graphene oxide (GO) to prepare O-HACC/PVA/GO dual self-healing bacteriostatic hydrogels. Then the hydrogels and materials were characterized by FT-IR, X-RD, 1H NMR, SEM and TG. The hydrogel's compressive strength, equilibrium swelling and bacteriostatic efficiency were systematically studied. The research results showed that the maximum equilibrium swelling rate of hydrogel was 720%, the maximum compressive strength was 1500 Pa, and could self-heal within 12 h. In addition, the hydrogel could effectively inhibit E. coli and S. aureus, and also showed a good release behavior for bovine serum albumin (BSA). The CCK-8 method proved that the hydrogel was non-toxic to murine fibroblasts and could promote cell proliferation and growth to a certain extent. This research has potential significance for the application of self-healing hydrogel materials in the field of biomedicine.
