项目标书摘要:日本脑炎(Japanese encephalitis)是由Flavivirus所引起,猪被认为是日本脑炎重要增幅动物,每年卫生署都会监测猪群血清日本脑炎抗体,并以转阳率超过百分之五十来发布人日本脑炎即将流行,但缺乏实际从猪只监测或病例分离本病毒的报告;亦未见其基因序列登录於NCBI Genbank。鉴此严重不足本计划拟以连续性之年度持续监测,采集猪只血清完成检测并建立台湾地区猪群血中出现日本脑炎病毒阳性比率是否呈季节消长形成完整循环周期。持续以高敏感性及特异性之即时定量RT-PCR方式直接检测病毒,监测台湾地区猪群不同季节猪只血中日本脑炎病毒含量高低情形;重新评估猪只在台湾地区是否真正拌演日本脑炎增幅动物角色。另外并进行配合按月采血检测猪群血清中日本脑炎抗体阳性比率是否随季节消长,比较猪群血清抗体转阳性比率增加之时间点是否与病毒含量消长相关;又是否与人病例通报出现月份及病例数消长情形相关。并将收集台湾地区猪流产病例日本脑炎病毒,分析其基因及抗原性与人病毒之相似度,作为流行病学分析参考。建立台湾地区本土猪日本脑炎抗原决定阳性检体日本脑炎病毒外套膜蛋基因全长解E基因序列资料库;分析监测所得台湾地区本土猪日本脑炎各病毒之抗原决定E基因序列及其相互亲缘关系,并登录於NCBI Genbank提供作为国内外学术研究参考依据。将计划监测阳性之检体进行病毒分离,所得之本土猪日本脑炎病毒株经由生物资讯学抗原变异基因分析,及各别与疫苗产生抗体进行免疫抗原性分析,将结果作为日後疫苗病毒株挑选之参考。另外,以溶斑减少中和抗体试验法评估现行台湾猪用及人用施打之商用疫苗所产生的抗体是否足以阻止本土猪日本脑炎病毒株在猪群感染增殖,是否得以保护人不受本猪源病毒感染。并将完成以溶斑减少试验法及西方墨点法进行本土猪日本脑炎病毒株对於具抗病毒潜力药物:Dehydroepiandrosterone、aspirin、indomethacin、sodium salicylate、ribavirin、selenazole、suramin、pentosan polysulfate降低病毒增殖之效力评估,作为紧急临床治疗药物选用之参考。

Application Abstract: Japanese encephalitis virus(JEV),a member of the genes Flavivirus that belongs to the family Flavividase,is transmitted between animals by Culex mosquitoes with swine being the major natural host for transmission to human beings.The peak endemic months appeared about one month after the date of 50%seroconversion prevalence rate in pigs in Taiwan(Chang et al.,2002).Although the pig is considered to be the important amplifying host,little is known about its epidemiological properties on swine herd blood viral load in Taiwan.To appreciate the real situation of JEV viral load in pigs'blood throughout the year,the survey is conducted using real time RT-PCR,which will be serial collected from 2007 to 2010.This study will be initiated to show the whole picture of the seasonal prevalence of JEV viremia in pigs in Taiwan.Several vaccines have been developed to control Japanese encephalitis in humans and domestic animals.It is therefore important to examine the genetic diversity of JE virus strains in Taiwan.We will examine the full length of the viral envelope protein nucleotide sequence from newly Taiwanese swine JEV isolates and compare them with those of vaccine strains by direct sequencing of polymerase chain reaction(PCR)-amplified products.In this study,we will characterize the antigenic properties of wild-tpye Taiwanese isolates of swine JEV and compare with two human vaccine strains(Nakayama,Beijing-1)and three swine vaccine strains(m,at,YL)using plaque reduction neutralization test.We finally evaluate the antiviral effect on JEV Taiwanese swine isolates of the Dehydroepiandrosterone,aspirin,indomethacin,sodium salicylate,ribavirin,selenazole,suramin,pentosan polysulfate,which are currently approved or in trial for clinical use,against Japanese encephalitis virus.
