项目标书摘要:运用杂交瘤(hybridoma)技术,建立大鼠垂体促性腺激素(gonadotropic hormone GTH)细胞与大鼠脑神经胶质瘤融合的杂交瘤细胞株。利用GTH杂交瘤细胞株既能连续传代,又能合成分泌促性腺激素的特性,克隆后,模拟发情周期的环境变化,通过细胞化学(cytochemistry)方法,研究克隆细胞随着环境变化所发生的动态变化,为论证"脑垂体促性腺激素细胞动态变化假说"提供实验依据。另一方面,某些稀有但非常重要且结构十分复杂的生物活性物质,直接通过生化方法提纯会受到原材料来源的限制,利用基因工程手段获得的途径也存在重重障碍。利用杂交瘤技术有可能解决这一难题。将具有传代能力的瘤细胞与某种可合成分泌珍稀活性物质的细胞融合,制备杂交瘤,筛选克隆后,可以大量培养,加上适当的诱导剂,有可能使其在细胞培养液中大量分泌所需的珍稀活性物质。

Application Abstract: We have overviewed the hormone variety and observed carefully the vaginal smears in each stage of the rat’s oestrous cycle.The related overview and thesis have been accepted by the《PROGRESS IN VETERNARY MEDICINE》and will be published.At the same time we put the emphasis on the research on the induction of mutation of the rat neurospongioma cells’(C6 cells’)hypoxanthine-guaninephosphoribosyl transferase(HGPRT)absence.At first,we try to get the cells what we want by 8-azaguanine concentration gradient,but we fail,and repeat it besides ultraviolet irradiation and then still fail.Recently we have obtained one strain of C6 cells which can exist in 6.0μg/ml MNNG after they have lived in 500μg/ml 8-AG and can also exist in 20μg/ml 6-TG then.According to the orginal plan,wo ought to have finished the C6 cells’mutagenesis and the preparation of the hybridoma this year,but because the expeiriment was interrupted,we havn’t still got HGPRT-cells to this day,and so we have not carried out the preparation of the hybridoma yet.Now remaining still a part of research funds,we wish the reseach would continue if we are permitted to postpone.If we can prepare the GTH(gonadotropic hormone)hybridoma cell strain,we are going to provide the ideal cell model for the research on the hypothesis of the gonadotropic cell dynamic changing and will look for a new way to prepare rare bioactive compound with the hybridoma technique.
