Application Abstract: In recent years,the knowledge of microRNA(miRNA)showed that the influence of miRNA is versatile and powerful in a wide variety of diseases.Evidences proved that anti-miR is a useful tool in clinical treatment.Our preliminary data showed that anti-viral drug AZT could induces cell senescence and apoptosis in human ovarian cancer cells,and this function might partial be derived from the activation of miR23a.This project planned to examine the biological role of miR23a in human ovarian cancer cells,and reveal the downstream target genes.The expression level of miR23a will be elevated by mimic RNA or miR23a containing plasmid,cell viability and related cell biological functions change will be monitored to estimate the real of role of miR23a in human ovarian cancer.Report assay will be used to explore the downstream target genes of miR23a,and the protein level will be evaluated to confirm the relationship.Finally,the differential expression level of miR23a between normal and tumor ovarian tissues will be compared to estimate the importance of miR23a in vivo.This will be done only if the financial support is sufficient.
