肠道病毒感染性疾病是人类最常见的疾病也是引起儿童死亡的主要原因之一,发病率和致死率均居儿童病因的前茅,肠道病毒71型(enterovirus71,EV71)病毒主要是通过人群间的密切接触传播,流行多见于夏秋季,多发生于学龄前儿童,也偶尔有成人病例。通常情况下EV71感染引起手足口病(hand,foot and mouth disease,HFMD),但部分重症患者可引起神经系统症状,病程迅速。 本报告以杆状病毒为疫苗载体,通过生物信息学分析将EV71 VP1基因扩增后与杆状病毒全长的GP64基因融合构建重组BmNPV(家蚕核型多角体病毒,Bombyx mori nucleopolyhedron virus),从而在杆状病毒中表达EV71 VP1抗原,并分泌到细胞外。最后通过沙鼠动物模型验证其免疫效果。 EV71 is one of the major pathogens associated with human hand,foot,and mouth disease(HFMD)in pacific Asia.Hand,foot,and mouth disease is a common viral illness that usually affects children under five,especially infants.Some cases suffered from EV71 infection presented severe neurological manifestation and even death.The report tried to use the baculovirus as a vaccine carrier.EV71 VP1 gene was amplified and fused with full-length GP64 gene.Then,the recombinant protein was expressed in baculovirus expression system,and secreted to the outside of the cell.Finally,gerbils animal model was used to validate its immune effect.
